Radix Sophorae Tonkinensis is the dried root and rhizome of Sophora tonkinensis Gagnep. (Fam. Leguminosae). The drug is collected in autumn, removed from foreign matter, washed clean, and dried.

shan dou gen contains alkalois such as matrine, oxymatrine, anagyrine and methylcytisine, flavones such as sophoranone and sophoradin, etc..

Anti-tumor effect

Gastrogavage of water infusion or warm infusion of guang dou gen at the dosage 60g/kg for consecutive 10~21 days to mice had significant inhibitory effect on inoculated cervical cancer U14, it had an inhibitory effect of over 25% on sarcoma S180. Intraperitoneal injection of water extract of powder of shan dou gen at the dosage of 500mg/kg had a curative rate of over 60% on sarcoma and solid liver tumor of ascitic type in rats. At the dosage of 300mg/kg, it could delay the formation of ascites and prolong the survival time in rats with solid liver tumor of ascitic type.

Matrine isolated from guang dou gen showed effects on ehrlich carcinoma in mice in vivo and in vitro. Matrine and oxymatrine were both effective on S180. Other ingredients also had inhibitory effect on S180. Japan shan dou gen had significant effect on malignant tumors with less side effects.

Inhibiting dehydrogenase of leukaemia cells

shan dou gen at the concentration of 2g/ml could inhibit WBC dehydrogenase in patients with acute lymphatic leukemia and acute granulocytic leukemia, which indicated that it had inhibitory effect on leukaemia.

Elevating WBC

Intravenous or intramuscular injection of total Ku Shen alkaloids or intramuscular injection of matrine and oxymatrine could elevate WBC.

Anti-arrhythmia effect

Intraperitoneal or intramuscular injection of total alkaloids of shan dou gen could antagonize arrhythmia induced by various drugs in animals.

Anti-bacterial effect

Matrine solution of over 0.3% had good inhibitory effect on streptococcus B, staphylococcus aureus, shigella dysenteriae, bacillus coli, bacillus proteus and bacillus pyocyaneus, etc.. Total ku shen alkaloids had inhibitory and killing effect on tubercle bacillus, leprae, comma bacillus, dermatomyces and leptospira, etc..

Anti-asthmatic effect

Total ku shen alkaloids and oxymatrine had significant anti-asthmatic effect on asthma induced by histamine in guinea pigs.

Others

shan dou gen could excite reticuloendothelial system, pituitary-adrenal cortex system, antagonize various toxins and inhibit the secretions of salivary gland. It also had blood pressure lowering, anti-febrile, liver protecting and anti-ulcer effects.

Chronic active hepatitis

Shan Dou Gen Injection, intramuscular injection, 2ml, 1~2 times daily, 2 months as a course of treatment. 402 cases of chronic active hepatitis were treated, and 218 cases were markedly effective, 151 effective and 33 ineffective.

Leptospira

shan dou gen 15g, da qing ye 60g, sheng gan cao 15g. 12 cases of leptospira were treated, and 11 were cured, 1 ineffective.

Tonsillitis, faucitis

bei dou gen andqui zhen cao of equal amount were made into tablets, 0.5g each tablet. 2~4 tablets, tid. 16 cases of tonsillitis and 4 cases of faucitis were treated and all were cured after 3.15 days averagely.

Arrhythmia

Oral administration of dauricine, 300mg, tid was used to treat 41 cases of arrhythmia. 32 were markedly effective, 5 effective and 4 ineffective.

Bladder tumor

Guang Dou Gen Extracturm,.4 tablets (equal to 1.5g crude drug), tid. Or Guang Dou Gen Injection 4ml (each ml equal to 2g crude drug) for i.m. bid, combining other therapy. 53 cases of bladder tumor were treated, 2 were clinically cured, 33 markedly effective, 11 effective and 7 cases were ineffective.

Malignant mole

Total dauricine and dauricine A or B were used for intravenous drip to treat malignant mole, among 104 treated cases, 91 were cured, 1 ineffective and 12 ineffective.
